The play count SHOULD update after a successful sync with your iPhone and iTunes on your PC. Below is a picture of the shortcut setup in the iOS Shortcuts app so you can create it for testing. If others could try this shortcut on their own devices and play a song (from start to finish) to see if their play counts updated that would be extremely helpful and much appreciated. So based on this, to me the issue appears to be more with the Music app in iOS than a communication issue between my PC and iPhone and iTunes. Instead it showed the same play count as before.
After playing the test song all the way through I then replayed it and ran the custom shortcut again expecting the play count to show as increasing by 1. I tested by playing a song and checking it's number of plays (while it is playing) with the shortcut. This shortcut, when run, displays a notification with the play count of the currently playing song (in the Music app). On the iPhone that has the problem I created a shortcut to test play count. When I rate a song on my iPhone the rating syncs to iTunes so it seems as though other data is being synced fine from both iPhone and PC in both directions. Whenever I edit a song in iTunes (add lyrics, rating, other info) the new info syncs to my iPhone fine.
However, if I play the same song on my iPod Classic and sync with iTunes then it updates within iTunes as having been played. I have recently noticed (though it could have been going on for awhile) that when I play a song on my iPhone 11 Pro it does not update that song in iTunes as having been played when I sync the iPhone with iTunes. I have numerous playlists in my iTunes that rely on play count to determine what songs are in that playlist.
